The DMK-led front lost the 2016 Tamil Nadu Legislative Assembly election to the AIADMK under J. Jayalalithaa, which was re-elected to power. The DMK itself emerged as a strong opposition, winning the largest number of opposition seats in the Assembly's history, while its ally the Indian National Congress fared poorly.

Significance
The defeat kept the DMK in opposition for a second consecutive term, but the party's strong showing positioned M. K. Stalin as a credible leader-in-waiting and set the stage for the 2021 revival.
